This past year, we have been studying the distribution of the amyloid precursor protein (APP) of Alzheimer’s disease in the squid giant axon as well as in the spinal cord of lamprey eel. Interestingly, we have found that APP localizes to plaques on the surfaces of isolated squid axoplasmic organelles and distribute to the organelle/microtubule interface. In addition, we have demonstrated that APP is highly concentrated in the postsynaptic terminal of the vertebrate spinal cord through confocal microscopy. We are presently studying APP distribution at the ultrastructural level using a variety of techniques in electron microscopy.
